Evidence for the extracellular delivery of influenza NS1 protein

چکیده

We constructed a reporter influenza A/Puerto Rico/8/1934 virus expressing truncated 124aa N-terminal NS1 protein fused to luciferase sequence (NanoLuc) without signal peptide. The reproduction activity of the vector correlated well with luminescent in lysates infected cell cultures or mouse respiratory organ suspensions. Surprisingly, we found that enzymatic was present not only intracellular compartments but also culture supernatants as sera bronchiolar lavages mice. This fact allowed us formulate working hypothesis about extracellular delivery mechanism protein. To test this idea, conducted co-transfection experiments Vero cells different combinations plasmids encoding genomic segments and chimeric NS1-NanoLuc plasmid. emergence compartment promoted by formation ribonucleoprotein complex (RNP) from PB1, PB2, PA, NP proteins. Therefore, may be delivered together nascent RNP complexes during maturation particles.

Applying conserved peptides of NS1 Protein of avian influenza virus to differentiate infected from vaccinated chickens

Avian influenza (AI) is a highly contagious disease in poultry and outbreaks can have dramatic economic and health implications. For effective disease surveillance, rapid and sensitive assays are needed to detect antibodies against AI virus (AIV) proteins. In order to support eradication efforts of avian influenza (AI) infections in poultry, the implementation of “DIVA” vaccination strategies, ...

Cloning and Expression of Influenza H1N1 NS1 Protein in Escherichia Coli BL21

Background:Influenza virus is globally pathogenic and it is usually associated with zoonotic respiratory diseases. This virus has caused a number of pandemics with a high mortality rate. The non-structural (NS1) protein of influenza A viruses is a non-essential virulence factor that has multiple accessory functions during viral infection. This protein is highly conservative. It has been shown t...

The multifunctional NS1 protein of influenza A viruses.

The non-structural (NS1) protein of influenza A viruses is a non-essential virulence factor that has multiple accessory functions during viral infection. In recent years, the major role ascribed to NS1 has been its inhibition of host immune responses, especially the limitation of both interferon (IFN) production and the antiviral effects of IFN-induced proteins, such as dsRNA-dependent protein ...

Human interactome of the influenza B virus NS1 protein

NS1 proteins of influenza A and B viruses share limited sequence homology, yet both are potent manipulators of host cell processes, particularly interferon (IFN) induction. Although many cellular partners are reported for A/NS1, only a few (e.g. PKR and ISG15) have been identified for B/NS1. Here, affinity-purification and mass spectrometry were used to expand the known host interactome of B/NS...

Conformational plasticity of the influenza A virus NS1 protein.

During infection, the influenza A virus non-structural protein 1 (NS1) interacts with a diverse range of viral and cellular factors to antagonize host antiviral defences and promote viral replication. Here, I review the structural basis for some of these functions and discuss the emerging view that NS1 cannot simply be regarded as a 'static' protein with a single structure. Rather, the dynamic ...
